Chromium Effects on Glucose Tolerance and Insulin Sensitivity in Persons at Risk for Diabetes Mellitus

This rigorously designed, randomized, double-blind, placebo-controlled crossover trial enrolled 59 adults with impaired fasting glucose, impaired glucose tolerance, or metabolic syndrome — precisely the population where chromium might be expected to work — and tested 500 mcg and 1,000 mcg of chromium picolinate daily for 6 months. Neither dose produced any significant changes in fasting plasma glucose, 2-hour glucose during oral glucose tolerance testing, fasting or 2-hour insulin, HOMA-IR, HbA1c, body weight, waist circumference, BMI, or blood pressure compared to placebo. The authors concluded that chromium supplementation does not ameliorate insulin resistance or impaired glucose metabolism and is unlikely to attenuate diabetes risk — an important finding directly relevant to this supplement's blood sugar and weight loss claims.

Satiety Effect of a Saffron Extract in Mildly Overweight Women

This double-blind, placebo-controlled RCT in 60 mildly overweight healthy women administered 176.5 mg of standardized saffron extract or placebo twice daily for 8 weeks. The saffron group showed a significantly greater reduction in snacking frequency (p<0.05) and a greater tendency toward satiety compared to placebo, alongside a significantly greater reduction in body weight. The proposed mechanism involves saffron's bioactive compound safranal acting on serotonergic pathways to reduce emotional/hedonic snacking — an effect on appetite behaviour rather than metabolism. This is one of the most-cited direct human trials supporting saffron for appetite balance, though it used a substantially higher dose than the 10 mg in this supplement.

Impact of Crocus sativus L. on Metabolic Profile in Patients with Diabetes Mellitus or Metabolic Syndrome: A Systematic Review

This PRISMA-guided systematic review of 14 RCTs specifically in patients with diabetes mellitus, metabolic syndrome, or prediabetes assessed saffron's effects on fasting blood glucose (FBG), HbA1c, and lipid markers. Of the higher-quality studies meeting strict dietary and activity assessment standards, three demonstrated significant reductions in FBG with saffron or crocin supplementation versus placebo. One study also showed a significant reduction in HbA1c. However, the review is candid that most available studies are of poor methodological quality, results on lipid profiles are inconsistent, and heterogeneity in doses (ranging from 15–100 mg/day) makes it difficult to draw firm conclusions. The authors conclude there may be a favorable effect of saffron on fasting blood glucose, but further high-quality research is needed.
