Oral Low-Molecular-Weight Collagen Peptide Improves Skin Hydration, Elasticity, and Wrinkling: A Randomized, Double-Blind, Placebo-Controlled Study

This rigorous 12-week RCT enrolled 64 women aged 40–60 with photoaged skin, randomized to receive either 1,000 mg/day of low-molecular-weight collagen peptide (LMWCP) or placebo. The collagen group showed significantly greater skin hydration as early as 6 weeks, and by 12 weeks, skin wrinkling and elasticity were also measurably improved compared to placebo — with the hydration improvement being 7-fold greater than placebo at 6 weeks. The proposed mechanism is that the tripeptide Gly-Pro-Hyp in the supplement is absorbed intact into the bloodstream, reaches dermal fibroblasts, and stimulates production of new collagen, elastic fibers, and hyaluronic acid, while simultaneously suppressing the matrix metalloproteinases (MMPs) that break collagen down.

Undenatured Type II Collagen for Knee Osteoarthritis: A Multicenter Randomized, Double-Blind, Placebo-Controlled Study

This 180-day multicenter RCT across 13 centers randomized 191 knee osteoarthritis patients to undenatured type II collagen (UC-II, 40 mg/day), glucosamine + chondroitin (1,500/1,200 mg/day), or placebo. UC-II produced significantly greater reductions in the WOMAC total score (a validated measure of OA pain, stiffness, and physical function) compared to both placebo and glucosamine-chondroitin, with significant improvements across all three WOMAC subscales. UC-II patients also required significantly less rescue pain medication than placebo — with only 11 UC-II patients needing it versus 28 in the placebo group. Notably, the glucosamine-chondroitin combination showed no statistically significant benefit over placebo in this trial.
